Mycotoxins are secondary fungal metabolites that are found frequently as natural contaminants in different foods that are habitual in the human and animal diet. They produce acute and chronic mycotoxicoses that affect mainly liver, kidney, and inmmunologic and reproductive systems. Moreover, some of them, as the aflatoxins (AFLs), have been recognized by the International Agency for Research on Cancer (IARC) as natural compounds having the highest teratogenic and carcinogenic potential for humans known, followed by ochratoxin A (OTA), fumonisins (FBs), deoxynivalenol (DON), zearalenone (ZEA) and patulin (PAT). The IARC has classified them into groups 1B, 2B, 2B, 3, 3 and 3, respectively. All of them are regulated in the highest risk food by the European Union. Mycotoxin-producing fungi are habitual saprophythes or parasites of agricultural crops all over the world, afecting mainly to cereals, fruits and nuts grown in warm regions, more favourable for their development. New scenarios associated to the regional and global climatic change are fostering new patterns of fungal distribution and dominance of the most competitive species under thermic and hydric stress conditions. This has exacerbated the problem of some mycotoxins produced by these species in agricultural crops. In this framework, new challenges in food safety and food quality related to fungi and emerging mycotoxins in food have been raised and it is necessary to tackle them. Recently, European competent institutions (EFSA, RASF, EC) have alerted the scientific community to the need of new research to know the identity, distribution, prevention, and sustainable control of the most relevant fungi and mycotoxins in food, considering as such, AFLs and OTA. Type A trichothecenes (T-2 and HT-2 toxins) have been summed up to them, owing to the high levels that have been found in cereals in Central and Northern Europe in recent years. Predictive studies related to the climate change locate Spain as a risky area with regard to fungi and mycotoxins occurring in cereals and grapes. Concerning the vineyard and wine, recent studies using advanced methodologies have permitted to establish that Aspergillus carbonarius and OTA, are dangers that are necessary to keep under control in agreement with our surrounding countries. Nevertheless, in spite of past studies, knowledge on this issue in cereals has become obsolete, and, consequently, the European Commission has demanded updated information to the member countries. Concerned about the current state of the topic, this Doctoral Thesis is aimed to the study of these risks related to fungi and emerging mycotoxins in agricultural Spanish crops. The research is organized in three chapters. The first chapter tackles the study of the mycobiota and mycotoxin contamination of Spanish barley in the main producing areas. This research is collected in articles 1 and 2 of the Thesis. The research starts with cereal sampling, which is representative of the main producing Spanish regions, followed by the detection, characterization and quantification of the fungal species in the collected samples. For this purpose, classical techniques based on the macro- and microscopic morphology of fungi, mycotoxin producing profiles and very updated species-specific PCR protocols have been used. Moreover, a comparative study of the two kind of methods, and a critical analysis of the advantages and disadvantages for its application in this field has been performed. This study has permitted to identify for the first time fungal species that had not been described previously in barley, such as A. carbonarius and A. steynii, two OTA producers. Moreover, A. flavus, the main producer of AFLB1 and AFLB2, has been detected as the most common mycotoxigenic species in the cereal. Together with them other species of great importance to food safety have been found. They are A. parasiticus, a well-known producer of AFLB1, AFLB2, AFLG1, and AFLG2, A. ochraceus and Aspergillus niger aggregate, which are OTA producers, Fusarium verticillioides, a producer of FB1 and FB2, as well as F. graminearum and F. culmorum, two species that can produce ZEA, DON and nivalenol (NIV). In addition to this, the species F. langsethiae, which can synthesize T-2 and HT-2 toxins, has been detected for the first time in Spanish barley as a result of the research. This species is characteristic of cereals growing in Central and Northern Europe and, so far, it had not been described in Spanish cereals. These results show new distribution patterns of mycotoxin-producing species in barley in Spain and, therefore, in Europe. In parallel with the mycological study of barley, this chapter deals with the study of the aforementioned mycotoxins in the cereal. With this purpose, analytical methods have been optimized and applied to mycotoxin extraction at high pressure, clean-up through different inmunoaffinity and solid-phase extration columns, and LC or GC analysis with different detection systems (fluorescence, refraction index, ultraviolet, electron capture or mass spectrometry) using pre- or post-column derivatization when required. This work, in addition to valuable methodological improvement, has allowed us to know that the most frequent mycotoxin in Spanish barley is OTA, followed by DON, AFLB1, ZEA, FBs, NIV, and T-2 and HT-2 toxins. Here the presence of AFLs in barley is described for the first time, although at levels below the established limits. Moreover, relevant information required by the EC on mycotoxin incidence in cereals in Europe is provided. The research performed in this first phase of the Thesis has been developed in collaboration with scientific experts of the Department of Genetics of the Complutense University of Madrid in the frame of coordinated projects funded by the Spanish “Ministerio de Economía y Competitividad (MINECO)”. The second chapter addresses the study of strategies for prevention and soustainable control of growth of the most relevant OTA, AFL, and T-2 and HT-2 producing species in the barley samples and the biosynthesis of the toxins. Research is aimed, first, at studying the most frequent ochratoxigenic species, A. carbonarius, which is also a well-known OTA producer in vineyards, and the species A. ochraceus, the major producer of this toxin in section Circumdati. Research continues with A. flavus and F. langsethiae, species producers of AFLs and T-2 + HT-2, respectively. Selection of fungi for this phase of the investigation is performed on the basis of the results obtained in the studied barley samples, prior studies carried out in grapes and the priorities of the European Commission in the area of mycotoxins. The research that deals with this second chapter of the Thesis is gathered in articles 3, 4 and 5, and very recent works, pending of publication that are also included and described in this chapter. Antifungal agents used in ecological agriculture (sulfur and copper oxychloride) and conventional agriculture (mancozeb) were assayed in the research carried out. Their effectiveness against isolates of A. carbonarius and A. ochraceus was acceptable. However, A. flavus and F. langsethiae showed a great resistance to these compounds and this required the assay of other most selective agents, such as azoles (prochloraz and tebuconazole) and a morpholine (fenpropimorph). In this Doctoral Thesis, for the first time and under different environmental conditions, the effective doses of all the assayed fungicides that yield fungal growth reduction of 50% (ED50) and 90% (ED90) are determined Their effectiveness to control OTA, AFLB1, AFLB2, T-2 and HT-2 production is also established. This makes it the first study on these antifungal compounds, fungi and mycotoxins performed under different humidity and temperature conditions expectable at short and medium term in the framework of the climate change. The results obtained are very useful for the design of good agricultural and storage practices and to alert from interactions between biotic factors (fungal species and strain) and abiotic elements (environmental factors, type and doses of fungicides) that affect the efficacy of the treatments on the control of fungal growth and mycotoxin production. Situations of medium stress (hydric, thermic, and low levels of fungicides) are detected and described for the different studied species, that far from controlling mycotoxin production, can enhance it. The research provides as well results of easy and inmediate transference to the business sectors involved in manufacturing and marketing of antifungals, as the companies Bayer CropScience, S.L., Industrias Afrasa, S.A. and Syngenta Agro, S.A., which have submitted all formulations used. A good part of this research has been performed in collaboration with scientific experts in the topic from the University of Cranfield (United Kingdom), a country with the highest level of contamination of cereals with toxins T-2 and HT-2, which strengthens its international scope. Finally, a third chapter is aimed at the study and design of strategies for OTA reduction in liquid media, which could be used in the technological processing of barley or grapes, e.g. in the manufacturing of beer or wine. The objective of this work was the search for alternatives to other possible, less innocuous and more agressive treatments, like the use of chemical adsorbents that can affect quality of the final product. This research is collected in articles 6 and 7 of the Thesis. For this research a species belonging to the lactic acid bacteria is chosen: Oenococcus oeni, a procariota that does not show toxic effects and responsible for the malolactic fermentation of wine. Ten O. oeni isolates are used, all of them obtained from red wine. The research is started in media favourable for bacterial growth supplemented with different concentrations of OTA (similar to those that can naturally be present in beer or wine). These media are innoculated either with bacteria at exponential phase of growth or heat-innactivated dead bacteria. In these cultures, toxin levels are monitored by LC with fluorescence detection. The experiences are also performed in media supplemented with different levels of ethanol to make them more similar to an alcoholic beverage like wine or beer. The overall results of both experiments are excellent, since depending on culture conditions, OTA reduction levels in the media ranged from 0,9% to 63%. The analysis of the results obtained permits to conclude that O. oeni presents ability to remove OTA from the medium, this process being independent of the viability of the bacteria, which indicates that O. oeni does not metabolize the toxin. The reduction is higher when the number of cells in the medium increases and concentration of the toxin in the medium decreases. The reduction rate is lower in media with high concentration of ethanol and very low pH. This agrees with the fact that OTA removal can be due to toxin adsorption to one or more components of O. oeni cell wall, probably the teicoic acids, and the pH of the medium influences the processes of adsorption/release of toxin to the medium and, consequently, the effectiveness of the process. The use of O. oeni as OTA adsorbent in alcoholic beverages, like wine, would be, thus, more effective in wines where malolactic fermentation occurs, since lactic acid is weaker than malic acid. This reduction process would be also favoured in beer and wine with low concentration of ethanol. This is the first study on the use of this species as a posible tool for OTA reduction in beverages derived from contaminated raw material. Further research applied directly to wine and beer, carried out with selected strains of O. oeni, will be a target for future work.Las micotoxinas son metabolitos secundarios fúngicos que se encuentran frecuentemente como contaminantes naturales en diversos alimentos habituales en la dieta humana y animal. Producen micotoxicosis agudas y crónicas que afectan principalmente al hígado, riñón y sistemas inmunológico y reproductor. Además, algunas de ellas como las aflatoxinas (AFLs) han sido reconocidas por la “International Agency for Research on Cancer” (IARC) como los compuestos naturales con mayor potencial teratogénico y cancerígeno para humanos que se conocen, a ellas le sigue la ocratoxina A (OTA), fumonisinas (FBs), deoxinivalenol (DON), zearalenona (ZEA), patulina (PAT) y citrinina (CIT). La IARC las ha clasificado en los grupos 1B, 2B, 2B, 3, 3, 3 y 3, respectivamente. Todas ellas están sometidas a legislación en los alimentos de mayor riesgo en la Unión Europea. Los hongos productores de micotoxinas son saprófitos o parásitos habituales en cosechas agrícolas en todo el mundo, especialmente, en las de cereales, frutas y frutos secos que se cultivan en regiones cálidas, más favorables para su desarrollo. Nuevos escenarios asociados al cambio climático regional y global están propiciando nuevos patrones de distribución fúngica y dominancia de las especies más competitivas bajo condiciones de estrés térmico e hídrico. Ello ha agudizado el problema de algunas micotoxinas producidas por esas especies en cosechas agrícolas. En este marco, se han planteado nuevos retos en seguridad y calidad alimentaria relacionados con hongos y micotoxinas emergentes en alimentos, que es necesario afrontar. Recientemente, las instituciones europeas competentes (EFSA, RASFF, CE) han alertado a la comunidad científica sobre la necesidad de nuevas investigaciones para conocer la identidad, incidencia, distribución, prevención y control sostenible de los hongos y micotoxinas más frecuentes en alimentos, considerando como tales las AFLs y la OTA. A ellas se han sumado los tricotecenos A (las toxinas T-2 y HT-2) debido a los altos niveles que se han registrado en cereales, en el centro y norte de Europa en los últimos años. Estudios predictivos relacionados con el cambio climático sitúan a España como zona de riesgo en materia de hongos y micotoxinas en cereales y uva. En el caso del viñedo y el vino, estudios recientes con metodologías avanzadas nos han permitido establecer que son Aspergillus carbonarius y la OTA, los peligros que es necesario controlar, y ello coincide con los países de nuestro entorno. Sin embargo, en cereales, a pesar de haber sido estudiados en el pasado, los conocimientos han quedado obsoletos por lo que la Comisión Europea demanda información actualizada a los países miembros. Conscientes del estado actual del tema, esta Tesis Doctoral se dirige al estudio de estos riesgos relacionados con hongos y micotoxinas emergentes en estos dos importante cultivos agrícolas españoles y al diseño de estrategias para su prevención y control sostenible. La investigación se recoge en tres capítulos. En el primer capítulo se aborda el estudio de la micobiota y micotoxinas contaminantes de la cebada española cultivada en las principales regiones productoras. Esta investigación se recoge en los artículos 1 y 2 de la Tesis. La investigación se inicia con un muestreo del cereal, representativo de las principales regiones productoras españolas, seguido de la detección, caracterización y cuantificación de las especies fúngicas en las muestras. Para ello se emplean técnicas clásicas basadas en la morfología macro- y microscópica del hongo, métodos cromatográficos aplicados al estudio de sus perfiles de producción de micotoxinas y protocolos muy recientes de PCR específicos de especie. Se realiza además por vez primera un estudio comparativo de ambos tipos de métodos, y un análisis crítico de las ventajas e inconvenientes para su aplicación en este campo. Esta investigación ha permitido identificar por vez primera especies que no habían sido descritas anteriormente en cebada como A. carbonarius y A. steynii productoras de OTA y se ha catalogado a A. flavus, productora de AFLB1 y AFLB2, como la especie micotoxigénica más frecuente en el cereal. Junto a ellas se han detectado otras de gran importancia en seguridad alimentaria como A. parasiticus, productora de AFLB1, AFLB2, AFLG1 y AFLG2, A. ochraceus y Aspergillus del agregado Niger, productoras de OTA, Fusarium verticillioides productora de FB1 y FB2, y F. graminearum y F. culmorum, productoras de ZEA, DON y nivalenol (NIV). Además de ello, se ha detectado por vez primera, en cebada española, la especie F. langsethiae, productora de las toxinas T-2 y HT-2. Esta especie es propia de cereales del centro y norte de Europa y hasta la fecha no había sido descrita en cereales españoles. Los resultados obtenidos muestran nuevos patrones de distribución de especies productoras de micotoxinas en cebada en España y por tanto, en Europa. Paralelamente al estudio micológico de la cebada, se aborda el estudio de todas las micotoxinas a las que se ha hecho referencia anteriormente en el cereal. Con este fin, se optimizan y aplican métodos para su extracción a alta presión, purificación mediante diferentes columnas de inmunoafinidad y de extracción en fase sólida, derivatización pre- o post-columna, cuando ello es posible, y análisis por LC o GC con diferentes sistemas de detección (fluorescencia, índice de refracción, ultravioleta, captura de electrones o espectrometría de masas). Este trabajo, además de aportar un importante avance metodológico ha permitido conocer que la micotoxina más frecuente en cebada española es la OTA seguida de DON, AFLB1, ZEA, FBs, NIV y las toxinas T-2 y HT-2. Aquí se describe por vez primera la presencia de AFLs en cebada, aunque a niveles que no superan los límites establecidos, y se proporciona información relevante requerida por la Comisión sobre incidencia de micotoxinas en cereales en España y por extensión, en Europa. La investigación recogida en esta primera fase de la Tesis se ha desarrollado en colaboración con científicos expertos del Departamento de Genética de la Universidad Complutense de Madrid en el ámbito de proyectos coordinados financiados por el Ministerio de Economía y Competitividad (MINECO). En el segundo capítulo, se aborda el estudio de estrategias para la prevención y control sostenible del crecimiento de las más importantes especies productoras de OTA, AFLs y las toxinas T-2 y HT-2, en la cebada, y de la biosíntesis de las toxinas. La investigación se dirige en primer lugar a la especie ocratoxigénica más frecuente, A. carbonarius, bien conocida también como productora de OTA en viñedos, y a la especie A. ochraceus como la mayor productora de la toxina de la sección Circumdati. Se continua con A. flavus y F. langsethiae, especies productoras de AFLs y de las toxinas T-2+HT-2 respectivamente. La selección de los hongos para esta fase de la investigación se realiza en base a los resultados obtenidos en las muestras de cebada estudiadas en el capítulo primero de la Tesis, a anteriores estudios realizados en uva y a las prioridades de la Comisión Europea en materia de micotoxinas. La investigación que conforma este segundo capítulo de la Tesis, se recoge en los artículos 3, 4 y 5, y en trabajos muy recientes, pendientes de publicación que también se describen en detalle en este capítulo. En esta investigación se estudia la eficacia de antifúngicos usados en agricultura ecológica (azufre y oxicloruro de cobre) y convencional (mancozeb). Su efectividad frente a aislados de A. carbonarius y A. ochraceus fue aceptable, sin embargo, la mayor resistencia de A. flavus y F. langsethiae a estos compuestos hizo necesario al ensayo de otros antifúngicos más selectivos como azoles (procloraz, tebuconazol) y morfolinas (fempropimorf). En esta Tesis, se determinan por vez primera, y en diferentes condiciones ambientales, las dosis efectivas de todos los fungicidas usados que reducen el crecimiento de las especies citadas en un 50% (ED50) y en un 90% (ED90). Su efecto en el control de la producción de OTA, AFLB1, AFLB2, T-2 y HT-2 fue también significativo. Ello le convierte en el primer estudio que se realiza con estos antifúngicos, especies fúngicas y toxinas en diferentes condiciones de humedad y temperatura esperables a corto y medio plazo en el contexto del cambio climático. Los resultados obtenidos son de gran utilidad para el diseño de unas buenas prácticas agrícolas y de almacenamiento, y alertan sobre interacciones de factores bióticos (especie y cepa fúngica) y abióticos (factores ambientales, tipo y dosis de fungicida) que afectan a la eficacia de los tratamientos en el control del crecimiento fúngico y producción de micotoxinas. Se detectan y describen también situaciones de mediano estrés (hídrico, térmico y niveles bajos de fungicidas) en las diferentes especies estudiadas que lejos de controlar la producción de micotoxinas por el hongo, la potencian. La investigación aporta, además, resultados de fácil e inmediata transferencia al sector empresarial implicado en la fabricación y comercialización de antifúngicos, como las empresas Bayer CropScience S.A., Industrias Afrasa S.A. y Syngenta S.A. que han colaborado y suministrado todas las formulaciones empleadas. Buena parte de esta investigación se ha realizado en colaboración con científicos expertos en el tema de la Universidad de Cranfield, (Reino Unido), país con mayor índice de contaminación de cereales con las toxinas T-2 y HT-2, lo que refuerza su alcance internacional. Finalmente, el tercer capítulo se dirige al estudio y diseño de estrategias para la reducción de la OTA en medios líquidos, susceptibles de empleo en el procesado tecnológico de la cebada o la uva, para la fabricación de la cerveza o el vino. El objetivo de esta investigación fue la búsqueda de alternativas a otros posibles tratamientos, menos inocuos y más agresivos, como el uso de adsorbentes químicos, que pueden afectar a la calidad del producto final. Esta investigación se recoge en los artículos 6 y 7 de la Tesis. Para esta investigación se elige una especie bacteriana del ácido láctico, Oenococcus oeni, un procariota responsable de la fermentación maloláctica del vino que carece de efectos tóxicos para la salud. Se usan 10 aislados de O. oeni, todos ellos obtenidos de vino tinto. La investigación se inicia en medios favorables para el crecimiento de la bacteria suplementados con diferentes niveles de OTA (similares a los que presenta la cerveza o el vino de forma natural) que se inoculan con O. oeni en fase exponencial de crecimiento o bien células muertas inactivadas por calor. En estos cultivos se realiza un seguimiento de los niveles de la toxina en el medio mediante LC-FD. Las experiencias se completan con ensayos en medios suplementados con diferentes niveles de etanol más parecidos a una bebida alcohólica como el vino o la cerveza. Los resultados globales de ambos trabajos fueron excelentes ya que, en función de las condiciones del cultivo, los niveles de reducción de la OTA en el medio por O. oeni oscilaron entre 0,9 % y 63% y el proceso es independiente de la viabilidad de la bacteria por lo que O. oeni no actúa metabolizando la toxina. La reducción es mayor a mayor número de células en el medio y menor concentración de toxina. La reducción es menor en medios con elevada concentración de etanol y pH muy bajo. Ello concuerda con el hecho de que la eliminación de OTA se pueda producir debido a la adsorción de la toxina a uno o más componentes de la pared celular de O. oeni, probablemente a los ácidos teicoicos, y que el pH del medio influye en la adsorción o liberación de la toxina en el medio y, en consecuencia, en la efectividad del proceso. El uso de O. oeni como adsorbente de OTA en bebidas alcohólicas como el vino sería, por tanto, más eficaz en vinos en los que ocurre la fermentación maloláctica, ya que el ácido láctico es más débil que el málico y también se vería favorecido en cerveza y en vinos con menor concentración de etanol. Este es el primer trabajo que se realiza usando esta bacteria como posible herramienta para la reducción de la toxina en bebidas derivadas de materias primas contaminadas, abriendo así una importante línea de investigación en seguridad alimentaria. Nuevas investigaciones directamente en vino y cerveza, con cepas de O. oeni seleccionadas, serán objeto de futuros trabajos.
